Abstract
The genetic information contained in DNA must be read correctly into messenger RNA before functional proteins can be produced. Eukaryotes contain two spatially separated genomes, contained within the nucleus and the mitochondria, and the expression of both genomes is necessary for energy production and mitochondrial function. In this thesis, I have investigated the roles of translation initiation, RNA processing and the regulation of tRNA abundance on fidelity in these genomes and highlight the mechanisms regulating translational fidelity that ensure coordinated protein production in eukaryotic cells.
