Abstract

This thesis encompasses the medicinal chemistry of two classes of natural products: pulvinamidesand pleuromutilins. The pulvinamide section firstly details the synthesis, characterisation andbiological activity of the known natural products. The biosynthesis of pulvinamides was hypothesized to arise non-enzymatically. Exploring this hypothesis through the synthesis and HPLC-HRMSscreening of pulvinamide derivatives against a lichen extract led to the discovery of 35 novelpulvinamides. The pleuromutilin section details efforts to modify the diterpene core to improve hydrophilicity and metabolic stability. Sidechain derivatisation was also explored, leading to the discovery of novel antibiotic compounds with a potency comparable to the antibiotic valnemulin.
