Abstract

The genus Mesorhizobium carry unique mobile symbiosis islands which confers on its host the ability to fix nitrogen and form symbiosis with leguminous plants. The protein RdfS facilitates the excision of these symbiosis islands from the bacterial chromosome. In this thesis, the role of the DNA-binding protein RdfS, a tyrosine integrase recombination directionality factor, is investigated. Using molecular genetic, biophysical and structural techniques, we determine RdfS can also facilitate transfer of symbiosis islands. RdfS additionally forms helical filaments in crystal which we believe allows for the specificity of RdfS-DNA interactions, and the associated biological implications of RdfS activity are discussed.
