Abstract
The aim of this study was to evaluate the effect of 2,4 D on callus production from the red rice (Oryza sativa cv. Barak Cenana) embryo. Rice seeds were dehusked by hands, sterilized using 70% alcohol, 25% Clorox, and 5% Clorox, respectively. Sterilized-seeds were cultured in MS media with additional 1 ppm, 2 ppm, or 3 ppm 2,4 D. Each bottle contains three seeds and the experiment was done with three replications for every treatment. One hundred percent of embryo produced callus in all of those three types of media. All calluses were emerged 18 days after cultured and located at the base of shoot. They are friable calluses with white yellowish colour. After 35 days of culture, embryo in MS media with addition of 1 ppm 2,4 D produced the highest callus fresh weight. Fresh weight of the callus in MS media with addition of 1 ppm, 2 ppm, and 3
ppm 2,4 D were 241,9 ± 57,9; 171,8 ± 30,9; and 156,7 ± 26,3 mg/bottle, respectively. In the future, MS media with addition 1 ppm 2,4 D can be chosen to induce embryo derived callus from Barak Cenana red rice. Barak Cenana embryo derived callus induced by 2,4 D is potentially regenerated to red rice plant.
