Melatonin, a neurohormone secreted principally by the pineal gland, is proposed to regulate the circadian rhythm. However, the absolute bioavailability of melatonin is variable and low because of poor dissolution, low oral absorption and extensive first pass metabolism. This study aimed to address these issues by designing a melatonin-loaded surge-sustained pulsatile system using combinations of solid lipids to fabricate microparticles via a spray congealing method using the Buchi Encapsulator. A range of lipids was trialled and it was found that Gelucire 50/13 microparticles are suitable to improve the solubility and dissolution profile of melatonin.
