Development and evaluation of taste masked paediatric medicinal formulations

Abstract

The aim of this thesis was to develop and evaluate novel peroral taste-masked paediatric medicinal products. The thesis firstly reports on the clinical evaluation of a novel tramadol chocolate-based delivery system (CDS) for taste acceptance and comparative population pharmacokinetics against a compounded tramadol hydrochloride oral liquid as comparator. A validated high performance liquid chromatography assay was developed to quantify tramadol and O-desmethyltramadol in human plasma in the presence of seven potentially interfering drugs. The second section of the thesis reports on the development via Design of Experiments of a novel taste masked formulation of the bitter antibiotic flucloxacillin.
